Blot blot blot
I have been busy doing western blot lately. There's quite some optimisation work to do. Yesterday, I was trying to do another round of WB, with shorter blotting time as my protein size is rather small. I prepared SDS-PAGE gel, transfer, washing, blocking, antibody reaction... Everything went on nicely until just now when I realised I used the WRONG membrane! What happened was the PVDF membrane was taken away from the place where I last left it, and without reading the label carefully I took the remaining membrane roll which is nylon made (thinking it is PVDF) and used it for blotting. I should have read the label to confirm and not rely on my habitual assumption... WB is quite laborious but what to do? I have no choice but to repeat the experiment. (>o<)
