PCRing

My PCR keeps giving me problems, yet I don't know what has gone wrong. I have optimised the parameters and repeated them to confirm that it is working on both cell line and also biopsy cDNA. But since two days ago, when I tried to do the expression study on my biopsy samples using the optimised parameters, all the PCRs did not give me any product bands, not even my internal control which is a housekeeping gene. I have been trouble shooting this and that, and still don't know what has gone wrong. When I am expecting results, I am almost always unavoidably being let down; but when I'm least expecting anything, things always turn out to be very smooth. Is this the norm in a research life?

If this is so, I am in the situation of the former one.

research = learn how to accept failure, face it and overcome it!
